About This Protocol

IGF-1 LR3 is a synthetic analog of Insulin-like Growth Factor 1 with an arginine substitution at position 3 and an additional 13-amino acid extension at the N-terminus. These modifications reduce binding to IGF-1 binding proteins, extending its active half-life from minutes to hours compared to native IGF-1.

Extended Half-Life
Native IGF-1 has a half-life of minutes due to rapid binding by circulating IGF-1 binding proteins (IGFBPs). The Long R3 modification reduces IGFBP binding affinity by approximately 1000-fold, extending the half-life to 20-30 hours and significantly increasing free peptide availability.
Downstream of Growth Hormone
IGF-1 is the primary mediator of growth hormone's anabolic effects. GH stimulates hepatic IGF-1 production, which then acts on muscle, bone, and connective tissue to stimulate protein synthesis and cellular proliferation through the IGF-1R receptor.
Research Context
IGF-1 research has examined its roles in muscle protein synthesis, satellite cell activation, and tissue repair. Studies in aging populations have examined IGF-1's relationship with muscle mass maintenance and physical function.
Monitoring Requirements
